![38 A GENETIC ANALYSIS OF HISTONE PROTEIN SUBTYPES IN YEAST 2. YEAST CONTAINS HISTONE SUBTYPES 2.1. H2B Yeast contains two copies of the genes for each of the four core histones (15). In keeping with the convention in yeast genetics (35), we designate the genes coding for histones H2A, H2B, HTA and НТВ, respectively. The organization of the HTA and НТВ genes is shown in Figure 2.1. The HTAl -НТВ 1 genes are genetically unlinked to the HTA2-HTB2 pair (15). In both cases the genes are transcribed divergently [these features are also found for the gene pairs coding for histones H3 and H4 (Mitchell Smith, chapter 1)]. To determine the primary structure of histone H2B and to determine whether the genes were coding for different subtypes, the НТВ 1 and HTB2 genes were sequenced, and the protein structure was inferred from the DNA sequence (7). Figure 2.2 is a comparison of the nucleotide sequences and inferred protein sequences of HTBl and HTB2. These data show that yeast НТВ genes code for histone subtypes.
