![Gene Regulation During Development / 25 of chromosome 1. The scutella from ten-day-old seedlings were used in subsequent genetic analyses since the difference in ADH activity observed between the two inbreds is greatest at this point in development (Fig. 19). Detailed genetic and statistical analyses indicated that the altered develop¬ mental program of ADH-2 in R6-67 is due to a single gene with alleles exhibiting dominant-recessive inheritance [Lai and Scandalios, 1980]. At ten days, heterozygotes had an ADH activity level indistinguishable from that observed in W64A, suggesting that the rapid decrease in ADH-2 activity observed in W64A and in the Fi progeny is due to a trans-acting dominant allele at this locus. This regulatory gene exhibiting dominant-recessive in¬ heritance was designated Adrl (alcohol i/ehydrogenase-regulator) and is found to be unlinked to either Cat2 or Adh2 (Fig. 20) [Lai and Scandalios, 1980]. It should be noted here that, the identification of Carl and Adrl in R6-67 was merely fortuitous, because we have identified several other inbred lines 400| R6-67 X W64A 01 2345678 Days Post Germination Fig. 18. Segregation of scutellar ADH activities in progeny from crosses between W64A and R6-67. Scutella were isolated from ten-day-old seedlings. Each circle represents one scutellum and was plotted along the horizontal axis according to its level of ADH activity (units per mg protein).
