Cells responsi- ; ble for the graft-vs-host reaction (a T-lymphocyte dependent function) are resis tant to in vivo treatment with Cortisol, whether these cells reside in the thymus, spleen or marrow. On the contrary, cells in the spleen responding to Phytohäma gglutinin (PHA) are sensitive to in vivo Cortisol. This indicates that the splenic GvH cell is not identical with the splenic PHA cell. While the response of spleen cells to PHA is depressed by in vivo Cortisol, the response of marrow cells from cortisol-treated mice is enhanced when compared to the PHA response of marrow cells from untreated mice. Whole-body irradiation (with marrow shielded) prevents this increase in marrow PHA in cortisol-treated . mice, suggesting that some peripheral PHA-responsive cells do survive Cortisol and are chased into the marrow. This is in accordance with recent findings in the laboratory (J.J. Cohen) that marrow from cortisol-treated mice has increased T cell activity in a cooperative T-B cell response to SRBC. Finally, the response of various PHA-responsive cell populations to anti-0 serum was tested with various dilutions of anti-0. Spleen cell suspensions treated with 1:3 anti-0 had 70-85% of the PHA activity destroyed. The PHA response of normal marrow cells was unaffected by undiluted anti-0. The.enhanced PHA response of marrow from cortisol-treated mice was 50% diminished by 1:3 anti-0 leaving residual PHA responsiveness equivalent to that of normal marrow. These experiments indicate heterogeneity in PHA-responsive cells with regard to their sensitivity to anti-0. Spleen cells contain a large proportion of áñti-0 sensitive cells, but the marrow PHA ceils are resistant to anti-8.
