The steroid hormone 20-0H ecdysone was covalently linked to its chromosomal binding sites—via its receptor(s) — by in situ photocrosslinking of salivary glands. After fixation, the glands were squashed and photographed (top). Chromatin binding sites were detected by immunofluorescence microscopy (bottom) using anti-ecdysone antibodies and fluorescing second antibody.
