Experimental embryology has demonstrated the importance of the cytoplasm in early development of the invertebrate and amphibian eggs. On the other hand many experiments have shown that the cell nucleus plays an increasingly important role after the initial period of cleavage. The various theories which have been presented to explain this fact will be discussed. The role of the nucleus in wall morphogenesis will be discussed in two different cases, the alga Acetabularia and the sea urchin egg. Morphogenesis in the absence of the nucleus will be correlated with the synthesis of macro- molecules (nucleic acid and proteins) under the same conditions. The biological and biochemical significance of experiments made on lethal hybrids, lethal mutations and nuclear transfers will be examined. Finally, recent experiments on chromatin activation and cell fusion will be presented.
